Tombstone's characterization in this run is pretty much the only thing that's been done well thus far.
Someone new. A character that no one sees coming. Or Doctor Octopus. He used to want to be in charge of the mafia and he is pretty smart. It could be a new way for Otto give Spidey hell

I was thinking when reading the discussion on whether Kingpin hates Daredevil or Spider-Man more that it would be interesting to do a story where the Kingpin is actually taken down, and stays down for a few years.
It could be interesting to have some rotating new Kingpins and several gang wars.
I'd go with the Owl for a year, since he has the history, it would raise his profile and it could be a story engine years down the line.
Another set-up might be Hammerhead with someone else as the front.
And you could easily introduce new characters like Madriporian yakuza or something.
It could also be interesting to have a negative version of a good guy, like a cop or something.

Could all be very interesting, and if they were to go back to the "Queenpin" concept, I could see Carolyn Trainor, who took up the mantle of Doctor Octopus for a while in the 90s after Ock proper was killed by Kaine (and after saving Peter's life while admitting to a deep, profound respect for him, no less), going the "high-tech crime boss" route like she did during that era.
