In-Universe, Captain America, Iron Man, Thor, although i would put Fantastic Four as the icons of mainstream superheroics, in comparison to vigilantes(Spider-Man,DD) or outcasts(X-Men).
In real world, pre-MCU, it was Spider-Man, Hulk, Wolverine, with Thing falling to fourth position(before Wolverine, Thing was certainly the third most famous marvel hero). Now, i feel it's Iron Man, Spider-Man and Wolverine.

I would put Dr Doom, Red Skull and Magneto as the Trinity of Earth's villainy, but in a wide, cosmic scale, yes, Thanos is in top 3.

The more I think about it, the more I feel the answer really is Iron Man, Captain America, and Thor. Sure Spidey is the most popular overall but the big three is still these dudes here. Hell, in DC, there have been many times where Wonder Woman probably wasn't one of the three most popular characters so the Avengers trinity being the big 3 makes perfect sense to me.

Also, since the Aquaman movie made more money than the Wonder Woman movie, then shouldn't Superman, Batman, and Aquaman be DC's big three, as far as inflation goes anyway? :^)

I think basing the Marvel "trinity" off of DC is a mistake. Two different universes with different characters and dynamics. It will never be quite right because of that.
I don't think Marvel has a "trinity", it does it's own thing and forcing a supposed trinity feels inorganic. Especially as it's always based of Superman, Batman, and Wonder Woman with people trying to force Marvel characters into their very specific roles.

Are we talking in-universe or out? In, I could see Cap and co. being it. Out, I have trouble seeing how anyone can topple Spidey.

Especially considering how many successful supeheroes have come along ever since Superman, Batman, and Wonder Woman's successes in the 1940s.

I'm not even sure that Wonder Woman belongs in DC's Trinity. DC has had to continuously print Wonder Woman for copyright reasons, but it often hasn't been very successful. Green Lantern was arguably DC's big third for long stretches of time.
