They all get my vote.
The Kate Spencer Manhunter. Greatest and most underrated super-heroine ever. If you haven't reader her book(s), go find them and read them.
I like too many to possibly list them all, but off the top of my head, it's a tie between the Earth-2 Huntress and Power Girl.

Well Batwoman (Kate Kane) is the character that got me into reading comics regularly.
And Black Canary was the star of my favourite series ever (Birds Of Prey)
But honestly, when Wonder Women is written well, there's no one better.
Wonder Woman. A character of many contradictions, yet manages to remain a standard of the heroic ideal in the DCU. Power, compassion, fighting ability, integrity, mythic backstory, kick-ass outfit - she's the full package.

Hawkwoman/Hawkgirl is probably my favorite DC heroine, but I really like Black Canary, Wonder Woman, Power Girl, Mera, Stargirl and Manhunter a lot too.

Barbara Gordon. Awesome as Batgirl, awesome as Oracle.
I don't know about the best, but Shayera Thal is definitely my favorite. She started out as somewhat of a sidekick to Hawkman in the Silver Age, but she's quickly evolved over the years to be every bit as intriguing and recognizable as her male counterpart, to the point that in some mediums she's arguably even overtaken him in notoriety.

While I wouldn't necessarily call her the greatest, Kate is indeed one of the most underrated characters in DC's stable right now.
Well, Wonder Woman is kinda unbeatable there. She represents so much.
BUT, i do love Courtney Whitmore (Stargirl). She's AWESOME.
Wonder Woman used to be a character that I didn't care for. Now, I just f&#king love her. I have Azzarello and Renae De Liz to thank for that.

Cassandra Cain- She was Batgirl when I was growing up, so she'll always be my Batgirl. Waiting for her New 52 return, I hope they don't water her down.
Hawkgirl/ Kendra Saunders- Similar to Cass Cain, she'll always be my Hawkgirl. I always thought she was a better character than Shayera. No disrespect.
