It's not the best story, but it is very accessible, and an excellent entry point to 200+ issues.
I'm not sure there's really another good answer. Pretty much all of the great Spider-Man stories require at least some knowledge of the history of the character(s) and series up to that point. Kraven's Last Hunt or Death of Gwen Stacy don't work out of context of the larger Spider-Man story.

With some of the other stories, there also isn't an obvious follow-up. If someone likes the first few issues of Ultimate Spider-Man, they'll probably enjoy the next arc.

if someone likes the Night Gwen Stacy Died, should they check out Amazing Spider-Man #123? If they like Kraven's Last Hunt, they might be happy with The Child Within, although it hasn't been reprinted a lot, and builds heavily on established continuity.
    Sincerely,

Single issue: maybe Amazing #314, that's a good one. Plenty of great single-issue stories in the Michelinie/McFarlane run. Amazing #252 would be a cool one too, though maybe you need the context of the original Secret Wars (maybe you don't though). One of the things I hated about Slott's ridiculous EPIC stories was that there was never time for the equivalent of a #252 where Spidey just took in what happened and reacted like a real human character might.

Story arc: Coming Home (Amazing Spider-Man Vol. 2 #30-37)

Single issue:Amazing Spider-Man#9. (I could had choosen any of the Stan Lee and Steve Dikto or John Romita Sr story without counting with the story arcīs they made and would be a good choice as well. But from the best of the best stories by Stan Lee issue#9 is my favorite)
Story arc:If This Be My Destiny (Amazing Spider-Man #31-33),Kraven Last Hunt and Return of the Sinister Six.

This is an awsome story, but it's a 2 parter, you must read issue #230 for the complete thing.

It's probably a good jump-on point, since if you like it, there are some other books to check out (the rest of Stern's run on Spider-Man, Mantlo's simultaneous run on Spectacular Spider-Man, DeFalco's later run of ASM, which would lead one to Peter David's Spectacular Spider-Man.)
    Sincerely,
    Thomas Mets
