I voted for Ferris Bueller's day off. But it was a hard choice because I really love The Breakfast Club and Planes Trains and Automobiles.

For film he directed Uncle Buck. But seriously the dude has some amazing films to choose from.
The Breakfast Club for me followed (closely) by Ferris Bueller's Day Off and then National Lampoon's Vacation.

I wasn't even thinking Ferris Bueller when I created this thread. I was expecting a dead-heat between The Breakfast Club, Home Alone, and Christmas Vacation.

I voted, per OP's parameters, for National Lampoon's Vacation, but it should be noted that that is also Harold Ramis's film, and not just John Hughes. Ramis directed.
