Why not. I can easily see her play the villain in a comic book adaptation, technically that's kind of what Harley is anyway, co-lead or not. And it's not like Gaga has that much room to demand specific type of roles just because she got a bunch of nominations for A Star is Born. She's still relatively new as an actress who just happened to already be a celebrity prior to venturing into movies.

She won awards for House of Gucci too. She is only trying for awards-worthy roles because otherwise she doesn't really need to have a film career when she has a music career to fall back on. Hollywood sees her as a huge get for their films though because her fans turn up to boost the box office.

Also, Circe wouldn't be a lead or co-lead role. Harley would be, especially if it is a film based on Mad Love or Harleen, so that's the difference despite both being "villain" roles.
