I dont think it will. Sam's been trying to create his own identity since issue one. Even if Matt were to die, I think they both would rather Sam stay Blindspot and be his own person. He's not the Devil of Hells Kitchen, he's Blindspot of Chinatown.

As I said earlier, something similar already happened when BP took over for Matt after Shadowland. I don’t think they’d want to revisit that well so soon or at all when that gimmick has been being used so often in so many comics to less than warm receptions. But it should be noted that unlike the example you have there and the many others that’s happened in, Sam is at least his own character.
