While there are a number of top tier female scientists in the DCU (many of them already mentioned in this thread), I'm not sure there are any that have been overtly portrayed in the Lex Luthor/Niles Caulder/Prof. Haley/Mr. Terrific/Dr. Sivana "Top of the Field in Every Known Field" kind of way.

There was a push to establish Veronica Cale into that rarefied realm a few years back, but getting it to stick has been difficult.

Personally, I think Marvel's done a better job on this front.

She was a supporting character in Steve Orlando's Supergirl run, which I think is probably her most recent thing.

Baroness Paula von Gunther was a Nazi scientist who was the first host of the demon "Dark Angel" during John Byrne's run on Wonder Woman. Later, she ends up on Paradise Island, and invents the Purple Ray of Healing.

Pre-Crisis, Von Gunther's history remains the same, except no Dark Angel was involved. She was just a Nazi scientist who later still invents the Purple Ray of Healing. Having been reformed by Wonder Woman personally, von Gunther stays on as the Amazon's chief scientist, and gets to become one herself (ie. with enhanced strength, speed and stamina like the other Amazons). A side note: the Purple Ray was also capable of turning mortal women into Amazons, which was done to Donna Troy.

Two of my favorites are Shay Veritas and Karen Beecher/Bumblebee (who should get a push outside of Teen Titans stuff).

If you're including "creating gizmos that qualify as superpowers"
    That's pretty much her theme.

You know, that's a character DC needs back in its lineup.

Not sure if you would think this character would qualify or not, but as I understand it, the Wonder Comics imprint is part of DCU.

In the Wonder Twins maxi-series, two of the characters that were created were Filo Math, a Nobel prize winning genius, and his teen daughter, Polly Math, she is also a genius. Polly has since become a friend of Zan and Jayna, and both she and her father are working with the Wonder Twins in a Hall of Justice related entity.
