1) Wonder Woman: imo, she is the greatest blend of human and superhuman in comics. That she's not DC's #1 property is a testament to writers' failure to develop an adequate rogues gallery, and the '50s obsession with "putting women back where they belong"
2) Superman: no getting around it, he's the first and the greatest, and the source from which all others have flowed, with a rich history and supporting cast
3) Batman: if Superman created the genre, Bats defined it's parameters. He has one of the (arguably, THE) best looks in all superhero comics. His history, lore and supporting cast are as rich as Superman's, but he has the benefit of being more adaptable. He can fit into any human time, or level of technology.
4) Hawkman/woman: The Winged Wonders (especially in their Thanagarian versions) are what I call All-Purpose Superheroes. You can plug then into almost any kind of story, against almost any of antagonist, from common thugs, to mystic monsters, to invading aliens. On top of that, they share one of the only looks in all superhero comics that casts Batman's claim to The Best in doubt. I put them together because (IMO) they're at their best as a loving couple, with distinct and complementary capabilities.
5) Mr. Terrific (Michael Holt): this character is unrewarded brilliance. He doesn't rank higher with me only because he lacks the heritage of the others, and has not built a respectable rogues gallery of his own. To me, he's The Man Of Tomorrow For Our Times. IMO, he's what Siegle and Schuster might have created had they suspected where technology might take us.
Honorable Mentions:
Dr. Fate: awesome look, amazing powers, great base, plus lots of creepy Lovecraftian story potential. He lacks only for some of the " how does magic work" and "who is this person" stuff that defines his Marvel counter-part. Oh yeah, and making him a dead space god's sock puppet doesn't help.
Hourman (the original): like Dr. Fate, great look, plus powers that lend themselves to an All-Purpose Superhero take. Plus The Minute Men, a crowd-sourcing idea way ahead of its time. He loses out only for lack of any respectable Rogues Gallery.

1. Superman
Everyone knows him as the first and stuff, but I think his greatness is underrated. He's had so many stories and phases that are alike and different in interesting ways.
2. Katar Hol
The most underrated, imo. He's pretty much the best of science fiction heroes and crimefighters, and he's got the best partner of any hero.
3. Batman
Lives up to his popularity for sure. If all he did was smoke bomb and grappling gun he'd still be one of the coolest.
4. Captain Atom
Very cool and human character with a really great power set. The 80s reboot is one of my favorite DC runs.
5. Vril Dox
technically not a superhero but a DC protagonist with superhero roots. Very clever and engaging.
