It is supremacist that humans are terrified of demographic changes, just as it is in countries where whites are terrified of the birth rates of other races.
In the case of mutants, it is a simple natural fact if there is no mutant genocide, as seen in ORCHID calculations in House of X # 1.

If it wasn’t, the earth would, other than some obvious practical changes, have cultural continuity in the handover from Sapien to Superior. Now, even the study of human languages will be discouraged in favour of Krakoan once the E-gene activates.
It's not supremacist rhetoric, it's stating evolutionary and biological fact. Taking into account of the findings of the scientists of the MU have proved this statement to be true for decades now, I don't see the reason for alarm, unless many that heard Charles' message were already hostile towards mutants. Hell, the average flatscan in the MU are hostile to other flatscans for arbitrary reasons as current. The main reason organizations such as ORICHIS exists is due to the irrational fear of replacement and using that to justify creating weapons of genocide with much of said technology having been created by prominent figures in the MU (Richards, Stark, Brashear, etc.), which was brought to the world's attention by Dr. Bruce Banner/the Hulk. As a result of this irrational fear, flatscans use the various mutates (many of the Avengers, Spider-Man, Fantastic Four, etc.) as proxies, and most likely, humankind's final line of defense against the perceived aggression of mutants.
In other words: the idea of there being mutant supremacist rhetoric is just another excuse for hostile human supremacists to make some grand call to arms, which could very well be their final chance at staving off said "replacement" instead of allowing evolution and nature run their respective course.

Because Moira, Xavier and Magneto know that it's a lie. Mutants are not the future.

There was a part of the fanbase that adopted demagoguery when they thought the inhumans were taking over. That's why the inhumans served as a metaphor for immigrants and refugees. It showed how they learned nothing from reading X-Men. Marvel took advantage and made DoX, etc. In the real world, not all minorities work together, and it was reflected in the comics.
Mutants are the next step in evolution, so humans won't be replaced, but it's their next phase. Like how humans developed speech and thumbs.
It was always weird having Inhumans be a metaphor for immigration when they were a classist society. It came off more as colonization with them bringing disease to wipe out natives rather than indigenous Americans looking for a new home.
Interestingly, mutants weren't mentionned in the future Reed Richards scampered around with Nathaniel in Hickman's run on Fantastic Four.
But really, put those words in anyone IRL see how long it takes for them to be shafted as an extremist by everyone else...
Bottom line being, yes, of course it is supremacist language, regardless of the in-universe justification to it.

It’s more like humans will become mutants. It’s only “replace“ in the sense that millennials will inevitably replace boomers.
