He's new. Daisy and Yo-Yo are the only members of the team from the comics, AFAIK.
There is a character named Alphonso Mackenzie in the comics that was introduced in 1988, though based on the character page the only thing they have in common is the name, which makes him similar to Lance Hunter: both are named after characters from Marvel that have some kind of connection to SHIELD but beyond that, they are minor characters. The fact that people have to ask or assume they are original characters to Agents of SHIELD shows how minor they are in Marvel Comics as a whole.
