Since nobody's mentioned her, I'd go with the thrill-seeking Joystick, real name Janice Olivia Yanizeski, who debuted as an enemy of Ben Reilly in his days as the sole web-slinger of New York City before and after he formally assumed (or resumed) the Spider-Man identity, though perhaps better-known for her stint on the Thunderbolts back in the 2000s. And while she's more an enemy of She-Hulk, Titania (Mary MacPherran) developed a severe phobia of Spider-Man after being trounced by him in the original Secret Wars event, something I always thought could've led to more of a grudge, especially because of their common origin of being bullied outsiders whose initial reactions to gaining power were somewhat similar. Of course, Peter ultimately became a hero, while Titania chose to descend into villainy by becoming a bully not so different from her past tormentors.
The spider is always on the hunt.

Black Cat aside, the next most prominent I could recall, would be Princess Python, of first appearing in Amazing Spider-Man #22 (March 1965), whom although only encountered Spider-Man at least one other occasion (The Amazing Spider-Man Annual 1998), she, as a prominent member of the Circus of Crime, went on against bigger leagues, including the Avengers and Hulk.
Here's two that nobody will recall. The Spider-Man musical with the originally planned female spider villain till story changes happened and Swiss Miss from 'Turn Off the Dark'

Nobody mentioned Calypso so far.
She was one of the main characters in the first story arc of Spider-Man by Tod McFarlane.

There was one, I believe, called Blitz from a Web of Spider-Man arc... New Enforcers, I think, can't remember. She only lasted a few issues, though.
Wish they'd use some of these gals again... Joystick, Delilah and Paper Doll I'd love to see again. Or Carolyn Trainer and Stunner brought back.
