Thanos wasn't a All seeing all knowing entity. He relied on spies and alien alligences for knowledge, he just had a lot.
With Ultron having lock and key over all communications going from and coming into Earth technologically and him having the Mind Stone which Already has instantly contacted Thanos across the Galaxy Ultron would have a better ways and means to see what's coming at Earth than Thanos would of what's actually happened to Earth since Loki's last transmission.
I mean it's now realistic that Ultron could have a Planetary Wakanda sheild over all of Earth and a Pym paricle shrunk Vibranium armored and Infinity powered drones waiting in space to pop up after the Sheild bounces the initial invasion force and Thanos would have no idea it was their or what awaits him on land.
Again, there's no reason why Thanos wouldn't have the Tesseract, which would allow him to bypass any shielding and simply step out through a portal into any location he wished.
In theory, he could have the Power, Reality and Space Stones before arriving, step out through a portal directly outside the door of the Sanctum with Ebony Maw, Cull Obsidian, Corvus Glaive and Proxima Midnight in tow, smash his way into the Sanctum with the four Black Order members, take the Time Stone with force, then do whatever the hell he wants.
Honestly, if he uses even a modicum of common sense, even a prepped Ultron has no chance, because he'd have four Infinity Stones - Power, Reality, Time and Space - before even encountering anything to do with Ultron.
Plus there’s nothings stopping Thanos from just bombarding the place before he invades which cripples Ultron’s defense off the jump.
This is a scenario match, and Thanos' preferred modus operandi is to invade with either his army or shock troops and take the stones by direct force, presumably because he wouldn't want to accidentally kill more than half the population at once.
Also, appearing outside the Sanctum wouldn't prevent Strange from Sling Ringing out of there. Maw might interfere with that, but considering what he was doing in Thor Ragnarok in the middle of his Sanctum Sanctorum, that seems a good way to get Maw teleported into the septic tank.
Meanwhile, opening up with nukes is entirely in character for Ultron when there's no JARVIS around to block him, as is trying to destroy all his enemies at once with an indirect attack (like Sokovia).
3 years of making drones 24hrs a day 7 days a week from hundreds of factories across the entire planet means that Ultron wins the numbers game, and again these drones could/would be made with Vibranium and be Infinity powered and possibly Pym tech'd up.
Plus Ultron knows where Thanos is at any time via the Mind Stone, so Thanos will only see Ultron if Ultron allows it. He would detect and be able to personally attack Thanos via Mind Stone before Thanos attacks him in any way.
It's not so much the fodder, but how well Thanos and his lieutenants are gonna deal with being sucker punched with one to a thousand nuclear explosions smacking into them before they know anything is happening, followed by an army of Xth generation Vibranium bodies designed by a mind based off of Tony Stark with access to Wakandan tech. Imagine a Bleeding Edge suit that didn't have to decide room or resources for a squishy middle-aged man, amped with force fields and Vibranium absorption/redirection gimmicks, times a few million.
Thanos is supposed to have the largest army in the galaxy behind him and equipped with equipment far more technologically advanced than Earth ever will be. We already know he has stuff that cuts through virbanium like a knife cuts through butter so it’s hardly an issue.
And where exactly are these on screen feats that allow Ultron to become an omnipotent god bot? Because I don’t recall seeing the mind stone doing anything close to what you’re suggesting.
All of which is irrelevant when Thanos can make it disappear with nothing more than a thought

No, but he does have a fleet in orbit so that should give him more than enough time to activate the gauntlet. That, of course, assumes that said fleet doesn’t shoot those missiles out of the sky long before they’re a threat to Thanos.
1 & 2) He used the gauntlet to wipe out 50% of all living creatures throughout the entire universe, I’m fairly confident the thing has more than enough juice to handle a measly nuke. If you want a feat specific to the reality stone, well he did transform Titan from a graveyard into a thriving environment (living people included)
Hell, with the reality stone Thanos can simply create a decoy than kick back and watch as Ultron’s arsenal does all the hard work of destroying everything before he ever sets a foot on earth.
