Tony Stark and T'challa have the skills and aren't cursed with the villainous tendency to trip over their own egos like Doom. Hydra/AIM, Namor or the like.
I could see Ultimate Reed Richards but most other Reeds are too science focused to handle spycraft manipulation and unless mutants are involved Magneto or X-Men aren't interested.
Loki would be a dark horse due to the lack of magic and flamboyance which are his trademarks.
Thanos isn't subtle enough.
We lack the violet shading that would indicate the Purple Man- plus even if Bendis could get him in do we really think DC would allow this story to center on Jessica Jones (or Bendis as calls her now, Lois)?
