In theory yes. In practice no.
Karnak should be a beast on paper with his abilities plus his slight meta human stats. But he apaprently is only effective against walls and heavy doors. It's a shame, as he could be a cool character if he were written more effectively.

This may be pushing the limit here...but Spider-Man. Combine his physical abilities, spider sense and the fact that he was trained in his own martial art..The Way of The Spider...by Shang Chi.
Though I wouldn't say he's the best... I think he's VERY underrated.
Very few people as fast and agile as him are as strong. Really no one who is as strong (or stronger) is as fast and or as agile. Add the spider senses (which compensates for the skill difference better fights have over him) and you've got one of the more effective h2h fighters in comics. He's not the most skilled by any stretch... but the unique combination of speed, strength, and spider senses makes him a handful for everything from a hand ninja to a god and everything in between.
