"Created Equal" though he's not the main character. A space virus passes Earth and kills the male population and only Superman survives (but a carrier for the virus). He leaves Earth in search for a cure but not before sharing how to combine Kryptonian and human DNA so a next generation of (hybrid) humans can be born.

It didn't had a virus or a disease in the classic fashion but it had many elements of an epidemic/pandemic situation.
And Superman played a significant part in this.
