The Killing Joke - It's considered a Batman classic for good reason I think.
"Of Thee I Sing"
(Hitman, Vol. 1 #34)
For me it's The Gauntlet. It shows Dick's "final exam"
The art by Weeks is amazing, and I love the building out of the mythos

Don't know if anyone will agree but I love this book.
If you like westerns, P&G probably had like 10 issues that could be nominees during their great Jonah Hex run (before All-Star Western). They specialized in one-and-done.
Hopefully this counts, but another favorite would be...
"Two of a Kind"
(Batman: Black and White, Vol. 1 #1)
“Dreadful Birthday, Dear Joker” by Len Wein and Walt Simonson, sometimes my favorite Joker story ever!
“The Adult Legion” by Curt Swan and Jim Shooter, Superman and the Legion!
“Fighters Never Quit” by Alfred Bester Paul Reinman, Golden Age Solomon Grundy vs Alan Scott GL!
Hopefully this counts, but another favorite would be...
"A Black and White World"
(Batman: Black and White, Vol. 1 #2)
Adventure Comics #452
A dark as hell tale considering it was released in 1977!

"My name is Wally West. I'm the fastest man alive!"
I'll try being nicer if you try being smarter.
Two Flash issues also spring to mind....
