With the right hook a LoD title could work. The Superior Foes of Spider-Man managed to be a critical darling, it's not beyond the LoD to at least churn out a decent run.
You wouldn't even necessarily need them to deal with "even evil-er" threats. Handled correctly, with the right goals, you could put the LoD against heroes. As the stars of the book the LoD has to eek out something resembling a victory (Aat least in most arcs), but that doesn't *have* to mean dead heroes or world tyranny either.
As for a roster....I think it'd be more important to get some interesting, quality villains with flexible goals, rather than just grabbing each Leaguer's archnemesis and throwing them into a group.
So perhaps something like.....

I like this idea. Especially if you play up the work place comedy aspect like JLU did. It would be interesting if you put them on Earth 3 and have them fight the CSA and try to rule that world.

I think it totally could be a book...people have said they'd have to fight other/worse villains, but who's to say they can't have victories over the heroes, too? Not everything has to be life/death or world-toppling...smaller victories can help sell the point too.
Anyway, my line-up. One rogue per gallery, per the rules, but still attempting to get to thirteen members. Going for something a bit different, a bit darker, more sinister and cerebral:

I love the original lineup, but there's a certain trapped in amber sense with them. These guys are always going to be around and they're never really going to change much. I think a new series should be a reinvention, the way Mystique reinvented the Brotherhood over on X-Men. I think I'd be more interested in a newer lineup where more unexpected things could happen seeing a bunch of villains we've never seen interact with one another.
