Realistically Gwen just to have a female playable character (I want Black Cat though) but they'd probably have to give her a new gimmick other than gliding (and maybe not the Symbiote since we're getting Venom).

Honestly, since they usually jam in many unique spider characters as costumes for Peter, (ie: Scarlet Spider and Spidey 2099) I never really feel the need for them to exist as individual characters. (I usually skip a lot of the cut scenes as well.) Unless they offer a two player simultaneous co-op mode, then Ben Reilly would make for a great player two "Luigi" to Peter's "Mario."
I guess a proper Symbiote Spider-Man would be cool. (Sort of like what was done in Web of Shadows.) MAYBE Spider-Gwen as a female avatar.(Although, I am getting a little tired of Gwen, personally.) Other than that I would rather play as a non-spider powered hero, such as the Black Cat or Daredevil. (Heroes with a similar way of traversing the city, but unique power sets.)

Well since Marvel wants to shove Jess into the Spider-Fam, Id say Spider-Woman Jessica Drew. Her play style would be pretty unique from the other spiders imo.

She got her powers from a serum made from irradiated spider-blood, even if said powers somewhat diverged from what we would normally think of as spidery. Believe it or not, there are some spiders that do indeed glide on air currents. That said, I do like the idea of Insomniac skipping ahead 20 years and giving us its take on Spider-Girl . . . or even Renew Your Vows. Personally, I'd also like to see them go further into the future and adapt Spider-Man 2099 for real, especially given how that era might look back on the existence of both Peter and Miles as Spider-Men.
