Or a life where he could be happy with the person he loved most with no complications or tragedies messing it all up.

As much as I could say Mephisto has gained a place in Spider-Man's foe gallery, I'm afraid that can't happen. I mean, Mephisto is a menace of cosmic level, while Spider-Man is just a "regular guy" (even with all his powers). The key for a good villain-hero rivalry is to make possible they can match each other's powers at some points, if not, it's an unfair difference with no point.

+ Superman's power can matches Darkseid's at some levels, and Lex Luthor can use kryptonite and use a battle armor.
+ Wonder Woman has the power of Gods and fights against Gods and titans.
+ Captain America is a super-soldier and fights against wannabee super-soldiers like Red Skull and Baron Zemo.
+ Iron Man uses tech to fight villain who use tech.

You get the idea. The Hero and Villain must have is similar conditions in order to even consider to be called "archenemies". Mephisto's power can only be match by someone like Doctor Strange, with great mystic power.

Mephisto is a metaphysical entity. He’s basically a Persona villain. It’s not about physical strength; it’s about strength of will.

Besides, villains are always at their weakest when they fight the heroes directly. In that case, it doesn’t actually matter how strong the villain is because that’s not how they do the most damage.

Persona/Spider-Man could be a cool crossover, too.

If they were to do a big story in which Spider-Man defeats Mephisto and restores the marriage, would anyone want a follow-up? This is one reason the arch-nemesis talk feels off.
    Sincerely,

This would be a big event and could tied into the current avengers run with the Mephisto war on the horizon

They already had one in Spectacular Spider-Man.

He can do more to affect the real world than the typical Persona villain, who typically need to gave groups of people on their side to have any real power.

Even then, the villains are defeated by people who have a supernatural power that can affect them, P3 protagonist sealed Nyx, P4 protagonist beat up the villain, P5 protagonist used a magic gun.

Spidey mostly doesn't get such power ups, having do defeat Mephisto is like asking a dog to defeat a tank.

So yeah, it's silly to expect Spidey to defeat Mephisto without questionable writing, which's why the end of Spencer's run, with either Spidey or Mayday being implied to be the one who defeats Mephisto, is real silly lol.

Only if Persona gets a game in USA, 'cause Spidey ain't really gonna travel to Japan for long, much less the year the games take place.

Spectacular comics got cancelled around two years after the first Persona game was made, and nobody cares about Persona 1, so I doubt even a reference happened, Spectacular's return didn't reference it too.

If you mean the cartoon, I don't remember character designs referencing it too.
