It seems "defining role" is really what the OP is after, less than "best" so much IMHO. Sometimes actors' "best" roles are little or lesser known films that never end up defining them.
James Bond is Sean Connery's best role, his defining role. (He's definitely my top Bond)
Daniel Radcliffe as Harry Potter, that defines Daniel.
Pacino may be defined by Michael Corleone.
Ray Liotta as Henry Hill in Goodfellas.
