The two kids, the Skrull N'kalla and the Kree Jo-Venn, adopted by The Thing,(Ben Grimm), and his wife Alicia Masters are two characters that were a good addition to the Fantastic Four family and also has long term potential.
Miles, Kamala and Spider-Gwen, for sure. Moon Girl is just under this.
In the 2000s, we had Runaways, Jessica Jones, The Sentry and Young Avengers
If it’s a hit, I’d keep an eye on characters in Hickmans upcoming “GODS” - if it’s got Hickmans name on it there’s a big chance it might stick.
Since Hickman ripped off from the inhumans to repurpose the mutants, post Disney throwing out the inhumans, I guess he's gonna take elements from Sandman for GODS. They probably have him the green light since Disney saw The Sandman series and wanted their own for the future at some point.
Disney knows branding matters, and having Sandman with a Marvel label on it will attract people.
It's obvious who are the most popular and successful, so I'm going personal faves
Mosaic
I like his powers, his character and really wish Redjack got more time with him.
Sam alexander and Kaldera
Novas and Champions won me over with Sam. Kaldera is fun villain for Sam to interact because she is just the silliest edge lord.
Twenty years is a a long time and so Ultimate Reed fits in, specifically as The Maker which started 2011
honorable mentions (no real solid justification I just want to see them return or stick around), Blinky, Auran, Rebecca ST. Jude, Lineage, progenitors, Alt universe Phyla, Reader, Iso

Miles, Gwen. X23, Kamala, and Squirrel Girl are the success stories. Moon Girl too
To a much lesser extent we got Maria Hill, Quake, and also does Ultimate Nick Fury count?
I'm going to throw in a vote for the Nuhumans as well, they got unfairly shafted. Some of them were pretty cool in my book.
Reading List (Super behind but reading them nonetheless):
DC: Currently figuring that out
Marvel: Read above
Image: Killadelphia, Nightmare Blog
Other: The Antagonist, Something is Killing the Children, Avatar: TLAB
Manga: My Hero Academia, MHA: Vigilanties, Soul Eater: the Perfect Edition, Berserk, Hunter X Hunter, Witch Hat Atelier, Kaiju No. 8
Kind of surprised at the interest for the new Inhumans. But it counts of course!
Don't know why I didn't think of Sentry!
Young Avengers-let'ss ee that was- Iron Lad, Hulking,Wiccan,Speed,Kate Bishop,Patriot/Elijah Bradley and Stature.
Does Iron Lad count? Wasn't he just a young Kang?
Later there was America Chavez ,Prodigy,Noh-Varr and ...well,Kid Loki if you want to count him. lol
Who is left standing if we disqualify copycats, analogues, alternate reality versions, family-based derivative characters, homages, etc.? Who is the best truly original character created in the last 20 years?

I know Moon Girl is technically a replacement for poor Moon Boy, but she has so little in common with him that I have a hard time calling her a substitute.
Kid Omega is also quite successful. For some reason that is beyond me, writers tend to like that little shit and are wanting to use him.
I'd say the number one "really original" new character would be Jessica Jones.
Bringing back the old, killing the young: that's the Marvel way
Lineage is an Inhumans villain. Rebecca St. Jude is a Kamala rogue, her codename is Lockdown. Auran, Reader and Iso are all Nuhumans - Iso is basically New Attilan's Prime Minister. Alt-universe Phyla is a version of Phyla-Vell, Captain Marvel, who was in Guardians of the Galaxy in the two runs before the current one.
