If they were to do a sequel trilogy (chances are they won't) I feel like the next thing would be to do a Mayday movie. Timeskip 14/15 years later.

If we do have another one, hopefully that one composes the team of actual fan favorites and not something that looks like the background extras that got missed when picking a team.
Do something like TAS's Spider Wars, a la, "We have met the enemy, and it is us." In a nutshell, our arachnid adventurers have to go up against (a version of) one of their own that's broken very, very bad and is now on a Multiversal murder spree, either to conquer or annihilate reality itself.

Considering Spider-Carnage in TAS was using the technology that created the Spot to magnify the detonation of an incendiary bomb so that it would spread across realities until existence itself was wiped out . . . having him actually acquire the ability to move through dimensions himself would be orders of magnitude worse. I could even see something like Jet Li's The One (which I still say the Amazing Spider from the Identity Wars crossover with Hulk and Deadpool back in 2010 kind of ripped off), with the rogue Spider-Variant trying to kill off the others by absorbing their powers and lifeforces and thus make himself a Multiversal god.

A rematch with the other Miles Morales from Spider-Men 2 could work.
Maybe a story with more evil Spider-Men. Or Spider-Verse Civil War.

Yeah, that could work better. I like the idea of "the call is coming from inside the house," wherein it's not some external or outside force coming after them, but that the conflict is coming from within, which makes sense when you consider that not everyone connected to the metaphysical Web of Life and Destiny is necessarily a hero or even a good person. That being said, I could see it working out like DC's Dark Nights: Metal/Death Metal where a bunch of "evil" Spider-Variants come together and antagonize the "good" Spider-Variants in a bid to dominate the Multiverse.

The movies could use Morlun and the Inheritors as an external threat for the Spiders to unite against.
I'd rather go with Kraven the Hunter forming an alliance of his alternates and alternates of his family members to hunt Spiders across the Multiverse. Either that, or a kind of Sinister 666, led by Doctor Octopuses and Electros from other universes, as per the first Web-Warriors arc post-Secret Wars, that had an alliance of Electros that came up with a method to tap into the unique electromagnetic frequencies of the Multiverse through their own electromagnetic powers, albeit using captive Doctor Octopus alternates to make the science work.
