A reboot's convoluted at this point since we've had two fairly recent takes on Spider-Man's origin.
The only way to make it fresh might be to expand on what Peter Parker's like before he gets bitten by the spider, and before he learns about Uncle Ben, and maybe exploring others who are affected by the storyline (IE- using the material about Crusher Hogan from the Tangled Web one-shot.)
It's cynical, but a different take on the romance angle might be Peter realizing that the girl he's in love with isn't good enough for him. Though I don't see that going over well with female moviegoers.
An advantage of a reboot is that Sony/ Marvel/ whoever gets the rights isn't limited to the decisions made in the Raimi or Webb films, so there's more flexibility with the villains.
It's still too soon to do any origin story.
