Well, since you're on the DC Threads, I'll recommend some essential DC graphic novels:

    1. Watchmen by Alan Moore and Dave Gibbons

    2. Dark Knight Returns by Frank Miller

    3. Justice League Origin by Geoff Johns and Jim Lee

    4. Sandman Endless Nights by Neil Gaiman

    5. Kingdom Come by Mark Waid and Alex Ross

(That's a pretty good place to start)

If you just want great reads I'd recommend the following:

    Batman Year One
    Dark Knight Returns
    All-Star Superman
    Kingdom Come

Any character in particular you're looking for? Do you want dark or light or funny or detective etc. stories?

Go to a shop or to an online store and just browse covers and descriptions. Grab the first two or three things that really get your interest.

There's no real "first issue" or introduction to DC in general, that I would say is going to work for everyone. You've got to go with your interests. You may love Red Son but hate Sandman. You may love Hellblazer but be bored by All-Star Superman. So, go with your gut.
Patsy Walker on TV! Patsy Walker in new comics! Patsy Walker in your brain! And Jessica Jones is the new Nancy! (Oh, and read the Comics Cube.)

Comic stories are generally propelled by forward momentum. Writers know that new readers are always jumping in, so they tailor their stories such that anyone can read a story arc without needing to know extensive background stories. If you want a few suggestions, the New 52 is a good starting point. Try starting with some of these runs:
    - Batman Vol. 1
    - Wonder Woman Vol. 1
    - Grayson Vol. 1
    - Action Comics Vol. 5
    - Green Arrow Vol. 4
    - Batwoman Vol. 1

For some older reads, try some of these graphic novels:
    - Superman: Birthright
    - All-Star Superman
    - Batman: The Long Halloween and Batman: Dark Victory
    - Batman: The Black Mirror
    - Kingdom Come
    - DC: The New Frontier
