Serious answer: Batman would probably go through some kind of existential crisis without the Joker, and he'll deal with this by coming down hard on said vigilante and accusing him of murrrderrr to project his own insecurities. Meanwhile, Gotham would be partying.
Batman would punch the killer in the face, bring him to the police, and then move on with his life.
Batman doesn't like Joker, but a murder is a murder.
And then Harley Quinn would become Joker.
His soul would evolve into a computer virus.
How Batman would react to the Joker's death varies widely with different interpretations of him. In the Dark Knight Returns, he spits on his face before setting his body on fire.

In Arkham City however, he carries Joker in his arms like he just lost his brother.
I'm sure he'll sleep a little easier at night not having to wait for the other shoe to drop waiting for the Joker to pop up again.
DC would come up with a character named The PUN or Bon Mot.
He watches Joker die in the DCAU as well, where he doesn't much care.
It's honestly a little hard to tell how he reacts to to Joker death in the DCAU. Not much is shown after Joker dies, and there's so much else going on in that scene (like Tim Drake having a psychological meltdown.) that it's hard to gauge what his reaction was. If I had to guess, I would say he wouldn't have cared much in this interpretation.
Maybe he would hang up the cowl and let Dick take over for good. End of an era with his main nemesis gone. Joker's been around since nearly the beginning.
Before the reboot there was a thread about which was the highest profile character not to die for real. I didn't think about it, I don't think any one did, but it was probably the Joker.

I remember BTAS where almost every episode with the Joker ended up with him being presumed dead, even if you do kill him off people would think the same thing of he's dead but is going to pop up again.
